  • We developed a simple method to produce haploids by the chemical treatment of pollen and identified ZmPOD65, which encodes a pollen-specific peroxidase, as a new gene controlling HI.
  • However, T2 progeny kernels of ZmPOD60-2 mutant heterozygotes had a relatively high abortion rate (4.3%–13.6%), which is in keeping with that of the ZmPOD65 mutant. These results suggest a key role of peroxidases in crop male sterility and HI.
  • After CRISPR knockout of ZmPOD65, offspring found an induction rate as high as 7.7%, proving that ZmPOD65 is a novel haploid induction gene.
  • A yellow fluorescent protein (YFP) fusion of one of these peroxidase proteins localized to the sperm cells in maize pollen grains , suggesting that ZmPOD65 ,ZmPOD60-1 and ZmPOD60-2 contribute to ROS defense in sperm.
