  • Taking the positional cloning approach, the Chr.9_ClpP5|clpp5|v30 gene, one of the proteolytic subunits of the chloroplast Clp protease complex, was identified and validated as the candidate gene for vyl.
  • The B73 Chr.9_ClpP5|clpp5|v30 protein consists of 281 amino acids, has a predicted chloroplast transit peptide at the N-terminal and a typical Ser–His–Asp catalytic triad.
  • Sequence analysis showed that there were multiple alternative splicing variants of the Chr.9_ClpP5|clpp5|v30 transcripts in the vyl mutant plants, all of which caused major changes in amino acid sequences and resulted in truncated proteins.
  • Disruption in vyl-modifier (Chr.1_Chr.9_ClpP5|clpp5|v30) alone does not result in any visible phenotype while loss of vyl (Chr.9_ClpP5|clpp5|v30) results in a chlorotic leaf phenotype and defects in chloroplast thylakoid stacking at seedling stage, suggesting the primary role of Chr.9_ClpP5|clpp5|v30.
